I'm an old newbie.
I used to read 2000ad way back in the mists of time then gave up as I set aside childish things.More fool me! Now i'm back and on my 14th prog.Its great to see that the progs grown up but still the same in humor and grossness. Talking of which Cradlegrave was awsome. I was really worried that after the complete fabness of Cradlegrave that I would find the comic sadly lacking but the one off tale this week was amazing. Great art i've never seen Tharg look so sexy!!
The only downside to coming in late or joining the prog now is that there are 3 tales with lots of backstory. Defoe, Red Seas and Sinister Dexter. Is there anyway I can find out the history to these? The one I find the most confusing is Sin Dex but all 3 are quite confusing.

Quick and dirty starter for 10 (I've found them especially handy for keeping the characters straight):

There are also trade paperbacks of the early stories (the Defoe one is imminent) and if you enjoy the stories then they are a good place to start.

I'm sure someone (like Dark Jimbo) will be able to fill in more and answer specific questions but...

The Sinister Dexter story is actually not so bad for jumping in on - they are guns for hire caught in the middle of a gang war. There are complications (as you'll have seen) involving a parallel universe but I think we are all still largely in the dark on some of this and if you hang on in I expect that future stories will start filling in the blanks and pulling  a swerves on us.
if I went 'round saying I was an Emperor just because some moistened bint had lobbed a scimitar at me, they'd put me away!
